ABSTRACT

Objective To learn the HIV infection status and related factors among spouses of HIV/AIDS. Methods We recruited HIV/AIDS cases diagnosed between January 2000 and August 2016 in Lu'an and their spouses to collect the demographic information and sexual behavior characteristics. For HIV sero-accordant spouses, we analyzed influencing factors for HIV infection by logistic regression model; while for HIV sero-discordant spouses, we followed up until December 31, 2016 and detected their HIV antibody periodically. Results The HIV infection rate of spouses was 20.35% (47/231) . Forty-five of the spouses had been infected with HIV before our investigation, with a infection rate of 19.48% (45/231) . Two of the spouses developed positive HIV antibody during the follow-up with a positive conversion rate of 0.39/100 person-years. Multivariate logistic regression analysis indicated that high frequency of sexual contacts before confirmed as HIV positive (OR≥1/week= 6.268, 95%CI:2.313-16.985; OR1/half a month=3.760, 95% CI: 1.268-11.150) and diagnosis during 2011-2014 (OR=3.079, 95% CI:1.209-7.841) were the risk factors for HIV transmission between couples; under the age of 25 (OR=0.042, 95% CI:0.004-0.467) when confirmed as HIV positive and early viral load<300 copies/mL (OR=0.115, 95%CI: 0.045-0.295) were protective factors. Conclusion HIV transmission of spouses is associated with frequency of sexual contacts, time of diagnosis, age when confirmed as HIV positive and early viral load.

ABSTRACT

The clinical data and ultrasound findings of 185 patients with thyroid nodules undergoing thyroidectomy from June to December 2015 were retrospectively reviewed.Postoperative pathology confirmed malignant thyroid nodules in 86 cases and benign nodules in 99 cases.The ultrasonographic findings showed that there were significant differences in obscure boundary [51% (44/86) vs.16% (16/99)],hypoecho [(95% (82/86) vs.27% (27/99)],internal micro-calcification [63% (54/86) vs.21% (21/99)] and lymph node enlargement [33% (28/86) vs.5% (5/99)] between malignant and benign nodules (all P < 0.05).The sensitivity,specificity,positive predictive value,negative predictive value and accuracy of thyroid imaging reporting and data system (TI-RADS) in differentiation of malignant and benign thyroid nodules were 77% (66/86),86% (85/99),83% (66/80),81% (85/105) and 82% (151/185),respectively.The positive rates of thyroid globulin antibody(TGAb) and thyroid peroxydase antibody (TPO-Ab) in malignant nodules [6% (5/79) and 25% (20/79)] were significantly higher than those in benign nodules [0 (0/94) and 12% (11/94)] (both P <0.05).There was no significant difference in free T3 (FT3),free T4 (FT4) and TSH between the two groups [(4.91-± 0.78) vs.(4.93-± 1.98) pmol/L,(13.57 ±2.22) vs.(13.58 ±2.17)pmol/Land(1.78±1.40) vs.(1.88±2.36)mU/L] (allP>0.05).The results indicate that TI-RADS grading and thyroid immunity index are valuable for differential diagnosis of thyroid nodules.

ABSTRACT

Abstract Patients with glycogen storage disease (GSD) are either orally fed (ORF) or gastronomy-tube fed (GTF) with cornstarch to maintain normal glucose levels. It is not known whether the use of cornstarch affects the microbiological oral profile of patients with GSD. Thus, the purpose of this study was to compare supragingival and subgingival plaque samples collected from 53 participants with GSD (2-56 years)—29 ORF and 24 GTF. The 16S sequence bacterial profiles of plaque DNA were obtained and a total of 768 probes were detected across the plaque groups. Orally fed patients showed higher means of cariogenic species and periodontal health-associated species, whereas GTF patients showed higher means of periopathogenic species (P < .05). Orally fed patients exhibited high levels of caries pathogens and lower levels of periodontal pathogens possibly due to the acidic environment created by their cornstarch diet, when compared to GTF patients.

ABSTRACT

<p><b>OBJECTIVE</b>To study the effect of vinorelbine on apoptosis, telomerase activity and expression of hTERT gene in human lung adenocarcinoma cell line Anip973 cells.</p><p><b>METHODS</b>Anip973 cells were cocultured with Vinorelbine at different concentrations and collected at 24 h, 48 h and 72 h after treatment, respectively. The inhibition rate of cell growth was determined by methyl thiazolyl tetrazolium (MTT) assay to evaluate the effect of vinorelbine. The percentage of apoptosis was detected by flow cytometry. The cellular morphology was observed under inverted microscope and electron microscope. Telomerase activity of Anip973 cells was determined by the method of TRAP-PAGE-silver staining. RT-PCR was used to detect the expression of hTERT mRNA.</p><p><b>RESULTS</b>Vinorelbine down-regulated the telomerase activity and expression of hTERT gene mRNA, inhibited the growth of Anip973 cells, and induced cell apoptosis in a time- and concentration-dependent manner. Annexin V-FITC assay showed 0.08 µg/ml NVB group could inhibited cell proliferation in 24 hour, and apoptosis rate was (7.37 ± 0.35)%, RT-PCR detection of hTERT mRNA expression in this group was (57.01 ± 1.71), and they were very significantly different from that in the control group (P < 0.01), but telomerase activity was (6.36 ± 0.06), compared with the control group showed no significant difference(P > 0.05). The change of hTERT mRNA expression is more sensitive than telomerase activity. The apoptosis rate, telomerase activity and hTERT mRNA expression of 0.4 µg/ml group and 2.0 µg/ml group were very significantly different from that in the control group (P < 0.01). The telomerase activity of 2.0 µg/ml at 72 hour group was (1.36 ± 0.27), basically completely inhibited, while the apoptosis rate was (74.87 ± 1.88)%, showed the cell apoptosis rate was in a negative correlation with the down-regulation of the hTERT mRNA (r = -0.96046, P < 0.01).</p><p><b>CONCLUSIONS</b>Vinorelbine can induce apoptosis in Anip973 cells, and its mechanism of action is related to telomerase activity. The detection of telomerase activity and expression of hTERT gene is useful in predicting prognosis of patients with lung adenocarcinoma.</p>
